We have 16 researchers in the academy, all have been working on this for most, if not all, of our careers. So that spans, for many of us, over 25 years. When we started, research on ageing at the molecular level was the backwater of biology, but in the last 25 years, ageing has moved to the forefront of science. It’s actually rare to open a leading scientific journal and not see a new breakthrough in our understanding of the ageing process.
There are two molecules that are special and find their way into the cell in a unique way, and as a result these molecules increased the endurence and lifespan in mice. NR and NMN are these molecules and they are both beneficial because they elevate levels of NAD+, which decline with age. NAD+ is vital to metabolism, turning nutrients into energy, and it also activates sirtuins, a set of genes that regulate longevity. We all possess NAD+ in our bodies thanks to our diet, by consuming foods with NAD+ precursors in them. While NR and NMN can be found in trace amounts in various foods, though, one can’t eat enough of anything to boost NAD+ levels back to youthful amounts. As a result, supplementing with a NAD+ precursor can help mitigate the decline. According to David Sinclair NMN is better than NR showing a higher more improved blood flow and endurance than NR.
Supplementing with NR and NMN could provide other myriad benefits as a result of boosting NAD+. NR and NMN supplementation in mice, for example, has proven beneficial for things like mitochondrial function. And NMN has shown improved blood flow and endurance as well as other age-associated physiological decline like low energy and weight gain. Additional research is needed to confirm similar effects in humans.
At the end of the day, though, what we know is that when compared to other precursors, NR and NMN are the most efficient of the bunch. The latest research however on NMN could prime it to be the next great generation, the new kid on the block.
NMN has shown to boost NAD+ levels and promote repair in damaged DNA, increases cell vitality helping to support cognitive functions., but for now, more research in humans is needed to better understand how beneficial it could be and how it might stack up against NR. One possibility is that each precursor could provide varying benefits depending on the target, especially if transporters, like the recently discovered one in the gut, are only available in specific cells in the body.

The generally recommended dosage of Nicotinamide Mononucleotide is anywhere between 50mg to1g per day. What is NMN?
NMN occurs naturally in the body and is found in dietary sources such as fruits, milk, and vegetables, however these dietary sources do not contain high enough amounts to keep your body at youthful amounts as you begin to age. NMN is used to generate nicotinamide adenine dinucleotide (NAD+), a powerful and essential molecule that is found in every cell of the body, this in turn activates a set of sirtuins genes that protect your body against disease.

What is Pure Trans Resveratrol?
Pure trans resveratrol is a resveratrol product that is at least 98-99 percent comprised of the active ingredient- trans-resveratrol. Resveratrol occurs naturally in such plants and plant products as dark chocolate, tea leaves, certain berries, grapes, peanuts and Japanese knotweed, which is then purified to various purities, anywhere from 5 percent to 99 percent pure. Pure trans resveratrol may cost more, but it contains effective dosages of the active ingredient, unlike many competitors, 5 to 10 percent pure, which are essentially useless.
Pure trans resveratrol has numerous health benefits, as mentioned above. These include protection against heart-attacks and strokes, anti-ageing and anti-inflammatory properties, , macular-degeneration and cataracts, and cancer-fighting properties.
In addition to trans resveratrol, low purity resveratrol supplements contain “cis resveratrol,” which likewise occurs naturally in the same plant products as does trans resveratrol. However, cis resveratrol has no known beneficial health properties. Most lower quality supplements contain large quantities of this useless cousin of trans resveratrol.

Niacin, also known as nicotinic acid, is an organic compound and a form of vitamin B₃, an essential human nutrient. It can be manufactured by plants and animals from the amino acid tryptophan.
Trans Resveratrol & NMN

Free radicals, antioxidants, DNA damage, and mutations, is incorrect, antioxidants have failed in the anti-ageing world.
Resveratrol is an antioxidant but doesn’t work like that in longevity, it steps on the accelerator pedal for sirtuins, so it’s directly controlling the bodies defences against ageing.

Anti-Ageing Pathway mTOR
A molecular complex called mammalian target of rapamycin (mTOR) is currently a major focus of pharmacological research to slow ageing, and for good reason. So far, reducing the expression of mTOR has been shown to extend lifespan across multiple species. Laboratory-bred mice whose expression of mTOR has been reduced to about 25% enjoy an approximate 20% increase in median lifespan.
mTOR is a cellular signalling pathway that serves as a central regulator for cell growth, metabolism, survival, and proliferation. This pathway is responsible for controlling many of the processes that use or generate large amounts of energy and nutrients. When mTOR signalling goes awry, it triggers numerous harmful events, including those linked to a variety of cancers.
Abnormal mTOR activation is a contributor to many of the chronic diseases of ageing. Scientists are aggressively seeking pharmaceutical approaches to lower the activity of mTOR.
Pterostilbene (as well as other CR mimetics and calorie restricted diets) has been found to naturally inhibit the mTOR pathway. This reduction in mTOR provides a powerful way of regulating cellular growth and metabolism, and combating some of the primary factors involved in ageing and disease.
pterostilbene impacts multiple antiaging factors involved in cardiovascular, hematologic, inflammatory, metabolic, and neurological disorders. Preclinical studies suggest that pterostilbene acts as a potent antitumor compound in multiple malignancies.
Cell and animal studies have shown that pterostilbene can extend the lifespan of various animal models of human longevity by regulating three major pathways linked to longevity: mTOR, AMPK, and Sirtuins. There are a lot of unknowns about which doors may open for these precursors in the future, and what they could open up to, which only makes the field of ageing research more exciting.
What is olive oil good for?
Olive Oil Contains Large Amounts of Antioxidants
These antioxidants are biologically active and reduce your risk of chronic diseases. They also fight inflammation and help protect your blood cholesterol from oxidation, two benefits that may lower your risk of heart disease. Dr. Gundry recommends cold-pressed, extra virgin high-polyphenol olive oil. He has not come out and recommended a specific brand, but in 2019 he did launch his own, Gundry MD high polyphenol olive oil.

Studies show that a high intake of animal protein, particularly red meat, which contains high levels of methionine and BCAAs, may be related to the promotion of age-related diseases. Therefore, a low animal protein diet, particularly a diet low in red meat, may provide health benefits. However, malnutrition, including sarcopenia/frailty due to inadequate protein intake, is harmful to longevity/metabolic health. Therefore, further study is necessary to elucidate the specific restriction levels of individual AAs that are most effective for longevity/metabolic health in humans.
Check your blood levels
When you start your journey on NMN, it's suggested you check your blood levels to check Methyl Donor Depletion, check your Homocysteine, Methionine and Betaine in blood, also your AST and ALT levels in your Liver. After 6 months taking NMN and Resveratrol, check your levels again to see if you have any change.
Normal levels of AST and ALT may slightly vary depending on the individual laboratory's reference values. Typically the range for normal AST is reported between 10 to 40 units per litre and ALT between 7 to 56 units per litre. Mild elevations are generally considered to be 2-3 times higher than the normal range.
Most laboratories report normal homocysteine levels in the blood between 4 and 15 micromoles/litre (µmol/L). Any measurement above 15 is considered high. Any measurement below 12 is considered low. Optimal homocysteine levels are below 10 to 12.
The elderly often have low methionine intake, and studies have shown that they may need higher intakes of 2 to 3 grams per day ( 24 , 26 ). Despite the fact that certain groups may benefit from increasing their methionine intake, many diets exceed 2 grams per day of methionine plus cysteine.
Resting concentrations of betaine in human serum range from ≈20 to 70 μmol/L with concentrations similar in diabetic subjects but lower in subjects with renal disease. High concentrations of betaine are found in human and animal neonates, probably because of choline metabolism
Choline, containing phospholipids and other substances, like glycerophosphocholines, are transported in blood lipoproteins. Blood plasma choline levels in healthy fasting adults is 7 - 20 micromoles per litre (µmol/l) and 10 µmol/l on average. Levels are regulated, but choline intake and deficiency alters these levels.
NR (Nicotinamide Riboside Side Effects)
Some people have reported mild to moderate side effects, such as nausea, fatigue, headaches, diarrhea, stomach discomfort and indigestion, if this is the case cut out high sugary sweet foods. In animals, taking 300 mg per kg of body weight (136 mg per pound) daily for 90 days had no harmful effects but they did not have high sugar in their diet.
Also the capsule itself maybe another reason behind why some people feel nausea and report indigestion, as this part although not made of plastic it does have that feel to it, it's unpleasant to swallow and can take time to digest.

This may be why turmeric intake has been associated with a reduced risk of age-related mental decline in humans. You can increase your curcumin intake by using turmeric in recipes or taking curcumin supplements.

It’s an integral component of your skin that helps maintain skin structure. As you age, collagen production slows, leading to collagen loss in the skin that accelerates signs of aging like wrinkles.

Coenzyme Q10 (CoQ10) is an antioxidant that your body produces. It plays essential roles in energy production and protects against cellular damage.
Research suggests that levels of CoQ10 decline as you age, and supplementing with it has been shown to improve certain aspects of health in older individuals.
For example, a study in 443 older adults demonstrated that supplementing with CoQ10 and selenium over 4 years improved their overall quality of life, reduced hospital visits, and slowed the deterioration of physical and mental performance.
CoQ10 supplements help reduce oxidative stress, a condition characterized by an accumulation of free radicals and other reactive molecules that accelerates the ageing process and onset of age-related disease. Though CoQ10 shows promise as an anti-ageing supplement, more evidence is needed before it can be recommended as a natural way to delay ageing.
Be sure to consult a trusted healthcare professional before giving it a try. CoQ10 is an important antioxidant that your body naturally produces. Some research shows that supplementing with it may slow age-related physical decline and improve quality of life in older adults.

Nicotinamide riboside (NR) and nicotinamide mononucleotide (NMN) are precursors to nicotinamide adenine dinucleotide (NAD+).
NAD+ is a compound found in every cell in your body and involved in many critical processes, including energy metabolism, DNA repair, and gene expression.
NAD+ levels decline with age, and this decline is thought to be associated with accelerated physical decline and the onset of age-related diseases like Alzheimer’s.
Animal studies have shown that supplementing with NAD+ precursors NMN and NR restores NAD+ levels and prevents age-related physical decline.

Crocin is a yellow carotenoid pigment in saffron, a popular, pricey spice that’s commonly used in Indian and Spanish cuisine.

Theanine. L-theanine is an amino acid concentrated in certain teas, including green tea. It may help protect against mental decline and has been shown to extend the lifespan of roundworms by around 5%.
Rhodiola. This medicinal plant has potent anti-inflammatory and anti-aging properties. One study in fruit flies demonstrated that treatment with Rhodiola rosea powder led to a 17% increase in their lifespan, on average.
Garlic. Garlic has powerful anti-inflammatory and antioxidant properties. Test-tube and rodent studies have shown that supplementing with this bulb may prevent UV-light-induced skin aging and wrinkles.
Astragalus. Astragalus membranaceus is a stress-reducing herb used in traditional Chinese medicine. It may help combat ageing by reducing oxidative stress, promoting immune function, and preventing cellular damage.
Fisetin. Fisetin tetrahydroxyflavone) is a dietary flavonoid found in various fruits (strawberries, apples, mangoes, persimmons, kiwis, and grapes), vegetables (tomatoes, onions, and cucumbers), nuts, and wine that has shown strong anti-inflammatory, anti-oxidant, anti-tumorigenic, anti-invasive, anti-angiogenic. Fisetin is a flavonoid compound that’s considered a senotherapeutic, meaning it can kill senescent cells. Blueberries are the primary dietary source of pterostilbene. Study after study has shown that consuming pterostilbene can prevent and mitigate age-related cognitive disorders, and higher intakes of blueberries have been found to reduce the rate of cognitive decline.
Pterostilbene is one of the most important contributing factors. Studies have shown that pterostilbene can improve cognition in rats. It has also been shown to help prevent the loss of the neurotransmitter dopamine from memory centers in aged rats. This is an example of how pterostilbene can battle the effects of age-related diseases.
In natural medicine, blueberry extract is said to have a number of health benefits. It is said to boost the health of blood vessels and is often used to treat or prevent the following conditions: Age-related macular degeneration. Alzheimer's disease.
